Preparation before sampling by a plankton sampler



1. The temperature and humidity of the operating room (clean room) under test shall meet the specified requirements; The static pressure difference, the number of air changes and the air velocity must be controlled within the specified value.



2. For the one-way flow test, the normal operation of the purification air conditioning system shall be no less than 10 minutes; The non-unidirectional flow shall start after no less than 30 minutes.



3. For static test, there are no more than 2 people in the room. During sampling, the tester should be downwind of the sampling port.



4. The tester shall follow the aseptic operation procedure. All measures shall be taken to prevent contamination of sampling ports and other man-made contamination of samples.



5. The tester must wear clean clothes that conform to the cleanliness level.



6. Before the phytoplankton test, the operating room (clean room) under test has been sterilized.







Sampling procedure of plankton sampler



1. Prepare petri dishes (XX clean room, XX measuring point) in a clean room with a platform, and wipe the platform with medical absorbent cotton soaked in 75% ethanol before operation. The tester's hands are rigorously disinfected in the same way. During the preparation process, the quality of the petri dish was carefully checked again, and it could not be used due to deterioration, damage or contamination.



2. Wipe the sampling port of the sampler, sample tube, the cover of the sampler, the turntable and the inner and outer surface of the cover with medical absorbent cotton soaked with 75% ethanol.



3. Do not put the sampler into the petri dish after disinfection. Start the instrument to run idling for no less than 5 minutes to make the residual disinfectant in the instrument evaporate.



4. Set the sampling amount according to the cleanliness level of the operating room. Flow rate (L/min)X sampling time (min)= sampling amount (L)



5. Put in the petri dish, cover and adjust the gap height of the sampler. When sampling, the petri dish should be handled with care and the outer diameter of the petri dish should be grasped with "five fingers" to avoid contamination of the sample.



6. Layout and number of sampling points.







What are the key points to note when using a phytoplankton sampler?



1. The instrument must be calibrated regularly according to the verification cycle of the test instrument. Use the instrument which has passed the test and is within the valid period.



2. If the test instrument does not enter the test area, clean the surface first or prepare and store in the corresponding clean room (protect the instrument with protective cover or other appropriate cover) if necessary.



3. When using paper in the class 100 clean room, a transparent dust free cover should be covered on it. Pencils and erasers are not allowed in the class 100 clean room.
