This experiment is a process of using a constant temperature and humidity incubator to induce germination of seeds under suitable temperature, moisture, and oxygen conditions, allowing seeds to swell and breathe, increasing enzyme activity, promoting metabolism, and improving germination vigor and germination rate. 1、 Select device capacity and temperature control accuracy
At present, the germination equipment put on the market includes wall insulated germination rooms and insulated quilt sets of soft sheds, with a seed storage capacity of thousands or more liters, which can be used for large seedling bases. A floor standing artificial climate box with fully computer-controlled temperature, humidity, lighting, and ventilation is used for germination, seedling cultivation, tissue culture, and periodic cultivation, suitable for laboratory use. The temperature control accuracy is expressed in terms of temperature deviation and its fluctuation. The digital temperature controller has higher accuracy, with a deviation and fluctuation of about ± 0.5 ℃.
2、 Pay attention to the humidity in the germination room
Due to low humidity, seeds are not easy to germinate, and high humidity makes them prone to rotting. Therefore, high-end climate boxes are equipped with humidity adjustment devices, and their relative humidity can be maintained between 60% and 85%. However, simple germination equipment does not have a humidity regulating device, and two measures are usually taken to ensure its humidity: one is to use an electric heating tube to directly boil water to produce steam and heat the germination chamber. The relative humidity changes little with temperature, and can generally be maintained at 60% to 85%.
